Design Notes

This DNA sequence was created by converting the amino acid sequence of RhNUDX1 using Genedesigner 2.0. We used the IDT Codon Optimizer tool to improve compatibility with E. coli, altering base pairs to preserve amino acid sequence. To remove restriction site problems with BsaI (due to the requirements of our DNA 2.0, our synthesis companies) we modified nucleotide 12 (G -> A) and nucleotide 33 (G -> A). A TAG stop codon was appended to the end of the gene.

Sequence found by converting translated amino acid sequence of RhNUDX1 protein (native to Rosa hybrid cultivar cultivar Papa Meilland breed roses) into the DNA sequence. The translated amino acid sequence is from GenBank entry: JQ820249.1, which gives the mRNA coding sequence of RhNUDX1 as well as the translated amino acids (also found as a protein at Genbank entry AFW17224).
